Tan Sri Datuk Seri Dr. Mazlan Bin Yusoff joined public service on 12 January 1988 as Pegawai Tadbir dan Diplomatik. He held senior roles which including as a Director-General of Malaysia Administrative Modernisation and Management Planning Unit (MAMPU) under Prime Minister's Department; and as a Director of Institut Tadbiran Awam Negara (INTAN).

Tan Sri Datuk Seri Dr. Mazlan Bin Yusoff is appointed as the Secretary General of the Ministry of Higher Education on 23 March 2020 and served until his retirement in late year 2021. During the tenure, he led efforts to modernise and elevate Malaysia's higher education institutions. He is also a champion of student welfare and institutional development. Under his leadership, numerous initiaties advanced the ministry's mandate. In November 2021, the Ministry publicly acknowledged his contribution with formal appreciation, followed by a dedication ceremony in his honour.

Tan Sri Datuk Seri Dr. Mazlan Bin Yusoff is appointed as Chairman of Education Service Commission (SPP) form year 2022 to January 2025.

During his post-retirement, he continued contributing as a respected figure in public sector leadership discourse. For instance, he delivered a talk at Universiti Utara Malaysia emphasizing the importance of visionary leadership, ethical conduct, and public trust in advancing national institutions.
